GLP-1 receptor agonists

What it is

GLP-1 receptor agonists are a class of prescription medications that mimic a gut hormone involved in appetite and blood-sugar regulation. Several branded products in this class are FDA-approved for diabetes and weight management. They are the most significant metabolic drugs of the past decade.

What is currently understood

Large, well-run human trials support their effects on blood sugar and body weight in the populations studied, under medical supervision. This is the strongest evidence base of anything in this library.

What is not established

Long-term outcomes in healthy, non-diabetic users are still being studied. Questions around lean-mass preservation, discontinuation and weight regain are active research areas, and the quality of compounded versions is a documented concern.

Regulatory status

Several are FDA-approved as branded prescription products. Compounded versions became widely available during the 2023 to 2025 shortage period; following resolution of those shortages the legal basis for compounding them is restricted and contested, and is an active enforcement area.
